While building my α10, I noticed that the instructions are slightly off. There are two STANDOFF4 1/2" #4-40 female - female in the parts list, but the instructions are written as if two more STANDOFF3 were used to attach the display to the LCDuino-1.

Thanks for the report. The instructions are fine, the parts list is what needed adjustment. There is no more "STANDOFF4" in the list. STANDOFF3 is used to mount the display module to the LCDuino-1 board as specified in the instructions. If you got two STANDOFF4 (female-female) standoffs, you could still use them for this purpose but instead of hex nuts, you'll need machine screws to fasten them to the display module.
